1536Sebastián de Belalcázar founds  the city of Cali, Colombia.
1540Captain Cristóbal de Oñate founded the city of Compostela, currently in Mexico, in New Spain, under the name of Santiago de Galicia de Compostela.
1540Juan Pérez de Guevara founds the city of Moyabamba, Peru.
1547The city of Guayaquil, currently in Ecuador, was founded in the viceroyalty of Peru by Francisco de Orellana, with the name Very Noble and Very Loyal city of Santiago de Guayaquil.
1553The city of Santiago del Estero, currently in Argentina, was founded in the viceroyalty of Peru by Francisco de Aguirre; it is the oldest city in the country still standing.
1567The city of Santiago de León de Caracas (Venezuela) was founded.
1577Alberto del Canto founded in New Spain the town of Saltillo, Mexico under the name of Santiago de Saltillo.
1693The city of Sabinas Hidalgo (Mexico) was founded in New Spain, under the name of Real de Santiago de las Sabinas.
1797The mission of San Miguel Arcángel was founded in New Spain, which will give rise to the city of San Miguel (California).
